Sterile Growth Factor Microspheres Enabled Cancer Vaccine Implant

Problem

A physician-sponsored clinical trial required a precisely dosed, sterile growth factor delivery system for incorporation into an implantable cancer vaccine scaffold. The challenge was to accurately load nano-gram quantities of a sensitive growth factor into biodegradable microspheres while meeting stringent sterility, bioburden, and endotoxin requirements for human use.

Solution

Phosphorex designed a PLGA microsphere formulation capable of reliably encapsulating nano-gram levels of the growth factor. An aseptic manufacturing process was developed to ensure sterility with low bioburden and endotoxin levels. We produced and released sterile microspheres under GLP conditions, supporting clinical use.

Outcome

The microspheres were successfully incorporated into cancer vaccine scaffolds at the clinical site and implanted in late-stage melanoma patients, enabling the execution of the physician-sponsored clinical trial.
